Vonoprazan is administered orally as a tablet and can be taken with or without food. The tablet should be swallowed whole with water and must not be crushed or chewed. It is typically taken once daily. No specific dose adjustments are required for mild to moderate renal or hepatic impairment, but caution and potential dose reductions are advised in severe cases. Precautions include monitoring for an increased risk of Clostridium difficile-associated diarrhea, potential bone fractures with long-term use, and severe hypomagnesemia. It is not indicated for pediatric populations. Elderly patients generally require no specific dose adjustments but should be monitored closely for adverse gastrointestinal effects and potential drug interactions, particularly with medications whose absorption is dependent on gastric pH.
